Abstract:

A major challenge in biological research is to integrate multiple levels of complexity, hierarchically ordered from molecules to molecular assemblies, to organelles, cells, tissues, organs and entire organisms. We have previously demonstrated that the small GTPase Rab5 is a master regulator of early endosome biogenesis. Rab5 is both necessary and sufficient (together with SNAREs) for the biogenesis of fusion-competent early endosomes in vitro for the entire endo-lysosomal pathway in vivo.

We are currently dissecting the molecular mechanisms whereby the Rab5 effectors mediate endosomal membrane tethering and fusion using a combination of protein structure, biochemical and biophysical approaches. At the cellular scale, we explored the design principles of the early endosomal network, which is shaped by a balance of fusion, fission and endosomal conversion (funnel model). The endosome tethering and fusion machinery responds to tyrosine kinase receptors (EGFR) to regulate their packaging in endosomes and determine the amplitude, lifetime and robustness of the signalling response.

We are now applying quantitative imaging and functional genomics approaches to explore the mechanisms underlying cell and tissue organization at the multi-scale level using the liver as model system.
